.Biogen deoligomerates bions after 6 days: implications for T cell-mediated immunotherapy.
The immunogenicity of biologics can be reduced by biogen deoligomerization, in which the biological entity is split into smaller, non-immunogenic, inactive entities. In this study, using BIAcore, we investigated the impact of bion deoligomerization on surface T cell epitope determinants and glycans on bion-specific bovine and human T cells. Deoligomerized bion surface presented an overall increased presentation of epitopes, compared with whole bion. Deoligomerization of a monomeric bion allowed the appearance of additional MHC class II- and class I-restricted epitopes, in both bovine and human systems. Additionally, deoligomerization of a bion allowed the identification of new N-glycan epitopes, in both bovine and human systems. We also detected a stronger proliferative response of bion-specific T cells when bions were deoligomerized. Our findings provide direct evidence that deoligomerization contributes to bion immunogenicity, and that bion deoligomerization should be considered to reduce the risk of an autoimmune response to therapeutic biologic products.Biological effects of interleukin-1β on cultured canine articular chondrocytes.
